The Routing Presets are a quick and easy way to assign audio streams to the analogue outputs on the back of the unit. You can change these manually by selecting audio streams yourself through clicking the drop-down boxes next to each output. The Routing Presets are just shortcuts.

DAW Tracking:

DAW Tracking assigns DAW 1 and DAW 2 to all analogue outputs in pairs. This means that the main outputs of your DAW, will pass audio to physical analogue outputs (including headphone outputs) on the back of the unit. DAW 1 and DAW 2 will also contain audio from the main system, if you choose for the main system output to be the Scarlett (so this will include audio from web browsers, media players etc.) This has many uses, one example is if you have multiple speaker setups connected to different outputs and want the same audio going to all of them (and they are many more uses too).

Zero Latency Tracking:

This is the default routing that your unit will use. It routes Mix 1 (L) and Mix 1 (R) to your analogue outputs in pairs. Mix 1 by default contains the main DAW outputs (DAW 1 and DAW 2) just as described in 'DAW Tracking' above, but it will also contain audio from your inputs as part of the Mix. This means you monitor your inputs directly, without latency. As the Mix also contains audio from the DAW, this is usually the best option to use when tracking, as it allows you to play along to previously recorded audio, with your direct (latency free) audio as well. Just remember to mute the channel you are recording in your DAW, so that you don't hear it twice! More information on latency can be found in this article.

Mixing:

This assigns each output from your DAW to the respective physical output on the back of the unit. So, if you assign something to output 7 in your DAW (for example), then it will come out of Line Output 7 on the back of the unit. This is good for the mixing process, as it allows for quick routing of channels from a DAW to outboard effects, or an external mixer.

Clear:

This just clears all routing of audio to the analogue outputs, meaning no audio is routed to them.
